Summary

Technetium-99m-labeled interleukin-8 ((99m)Tc-IL-8) effectively visualizes osteomyelitis lesions in rabbits. Despite lower uptake, its high target-to-background ratios suggest it may be a suitable agent for acute osteomyelitis diagnosis.

Area of Science:

  • Nuclear Medicine
  • Radiology
  • Infectious Disease Imaging

Background:

  • Early and accurate diagnosis of osteomyelitis is challenging.
  • Radiography is insensitive in early stages.
  • (99m)Tc-methylene diphosphonate (MDP) bone scans are sensitive but lack specificity.

Purpose of the Study:

  • To evaluate the performance of (99m)Tc-interleukin-8 (IL-8) in an experimental model of acute osteomyelitis.
  • To compare (99m)Tc-IL-8 with other imaging agents.

Main Methods:

  • Acute osteomyelitis was induced in rabbit femurs.
  • Animals were imaged using radiography, (111)In-granulocytes, (67)Ga-citrate, (99m)Tc-MDP, and (99m)Tc-IL-8.
  • Quantitative analysis included uptake and target-to-background ratios (T/Bs).

Related Experiment Videos

Main Results:

  • All agents detected osteomyelitis in rabbits where it developed.
  • (99m)Tc-IL-8 showed significantly higher T/Bs due to fast background clearance.
  • Radiography had low sensitivity (2/8 rabbits).

Conclusions:

  • (99m)Tc-IL-8 clearly revealed osteomyelitic lesions in a rabbit model.
  • Its favorable T/Bs, ease of preparation, and lower radiation burden suggest potential for acute osteomyelitis scintigraphy.
