Background:

  • Myelodysplastic syndromes (MDS) are a group of clonal hematopoietic stem cell disorders.
  • Accurate diagnosis and prognosis are crucial for effective management of MDS.

Purpose of the Study:

  • To evaluate the diagnostic and prognostic value of immunophenotyping in myelodysplastic syndromes (MDS).

Main Methods:

  • Analyzed surface markers on mononuclear bone marrow cells from 32 MDS patients using indirect immunofluorescence and monoclonal antibodies.
  • Correlated antigen expression with FAB subtypes and disease evolution.

Main Results:

  • Observed increased myeloid antigen and decreased lymphoid antigen expression in MDS patients.
  • Found elevated CD13+, CD33+ immature myeloid cells and decreased lymphoid antigens correlating with MDS progression (RA/RAS to RAEB/RAEB-t).
  • Increased CD34+ cells in RAEB/RAEB-t subtypes and elevated early marrow cell antigens (CD38, HLA-DR, CD99R, CD9) were noted. CD34 expression predicted transformation and poor survival.

Conclusions:

  • Surface marker analysis of bone marrow cells is a valuable tool for MDS diagnosis.
  • Immunophenotyping aids in predicting MDS prognosis and disease transformation.
