Changes in tight junctions of thyroid epithelium with changes in thyroid activity.

L W Tice, S H Wollman, R C Carter

    The Journal of Cell Biology
    |September 1, 1975

    Summary

Thyroid epithelial tight junctions appear normal under most conditions. However, thiouracil administration, a goitrogen, alters tight junction morphology, creating variations in width within rat thyroids.

    Area of Science:

    • Cell Biology
    • Endocrinology
    • Histology

    Background:

    • Tight junctions are crucial for maintaining epithelial cell polarity and regulating paracellular transport.
    • The specific morphology of tight junctions in thyroid epithelium and their response to physiological stimuli are not fully understood.

    Purpose of the Study:

    • To investigate the morphology of tight junctions in rat thyroid epithelium.
    • To determine how changes in thyroid activity and hormonal stimulation affect tight junction structure.

    Main Methods:

    • Freeze-fracture electron microscopy was used to examine glutaraldehyde-fixed and glycerinated rat thyroid tissue.
    • Morphological analysis of tight junctions was performed under normal conditions, after short-term activity changes, hypophysectomy, and thiouracil administration.

    Related Experiment Videos

    Main Results:

    • Normal rat thyroid tight junctions exhibit morphology similar to other epithelial cells.
    • Short-term thyroid activity changes and hypophysectomy did not significantly alter tight junction appearance.
    • Thiouracil treatment led to the formation of unusually narrow or wide tight junctions, and heterogeneous junctions within the same cell.

    Conclusions:

    • Rat thyroid epithelial tight junctions are generally stable but can be dynamically altered by specific hormonal stimuli.
    • Goitrogen-induced stimulation, via thyroid-stimulating hormone, impacts tight junction width and integrity.
