Summary

This study demonstrates that the kidney, liver, and lung can synthesize aldosterone, a crucial hormone. Aldosterone synthase gene expression was confirmed in these extraadrenal tissues.

Area of Science:

  • Endocrinology
  • Molecular Biology
  • Physiology

Background:

  • Aldosterone is primarily synthesized in the adrenal cortex.
  • The extraadrenal synthesis of aldosterone has been investigated in vascular and brain tissues.
  • Further exploration into other extraadrenal sites is warranted.

Purpose of the Study:

  • To investigate the potential of extraadrenal tissues, specifically the kidney, liver, and lung, to synthesize aldosterone.
  • To identify the expression of the aldosterone synthase gene (CYP11B2) in these tissues.

Main Methods:

  • Ex vivo kidney perfusion in Wistar rats (normal, ACEI-treated, adrenalectomized).
  • Analysis of perfusate for aldosterone using HPLC and RIA.
  • RT-PCR, Southern blot, and in situ hybridization to detect CYP11B2 mRNA expression in kidney, liver, and lung tissues and cells.

Related Experiment Videos

Main Results:

  • Aldosterone production was detected in kidney perfusate, unaffected by adrenalectomy.
  • CYP11B2 mRNA expression was confirmed in kidney tissue, cultured renal tubular epithelial cells, liver, and lung.
  • In situ hybridization localized CYP11B2 mRNA to liver fat-storing cells (Ito cells) and lung type II alveolar cells.

Conclusions:

  • The kidney, liver, and lung possess the capability to produce aldosterone.
  • These findings indicate significant extraadrenal sources of aldosterone synthesis.
