NG2 immunoreactivity on human brain endothelial cells.

Summary

NG2 is expressed on human brain endothelial cells, including non-proliferating cells and those in nonmalignant tissues. This finding expands our understanding of NG2 expression in the central nervous system (CNS).

Area of Science:

  • Neuroscience
  • Cell Biology
  • Vascular Biology

Background:

  • NG2/CSPG4 is a transmembrane proteoglycan expressed by oligodendrocyte precursor cells and pericytes in the central nervous system (CNS).
  • Its expression in brain endothelial cells, particularly in non-tumorous and non-proliferating contexts, is less understood.

Purpose of the Study:

  • To investigate the expression of NG2 on human brain endothelial cells.
  • To determine if NG2 expression is limited to proliferating endothelial cells or those in brain tumors.

Main Methods:

  • Dissociated cell cultures from human temporal lobe epilepsy resections.
  • Reverse transcription-polymerase chain reaction (RT-PCR) for mRNA analysis.
  • Immunocytochemistry and immunoblotting for protein analysis.

Related Experiment Videos

  • Immunoreaction on human cerebral microvessels from nonmalignant CNS tissues.
  • Main Results:

    • NG2 expression was detected at both mRNA and protein levels in human brain endothelial cells.
    • NG2 was found on both proliferating and non-proliferating endothelial cells.
    • Human cerebral microvessels in nonmalignant CNS tissues also showed NG2 immunoreaction.
    • Both anti-rodent NG2 and anti-human NG2 antibodies confirmed protein detection.

    Conclusions:

    • NG2 is expressed by human brain endothelial cells beyond proliferating cells and tumor-associated vasculature.
    • This study broadens the known cell types in the CNS expressing NG2.
    • Endothelial NG2 expression is not exclusive to pathological conditions like tumors or active proliferation.
