Localization of a thrombin-sensitive calcium pool in platelets.

T Sato, L Herman, J A Chandler

The Journal of Histochemistry and Cytochemistry : Official Journal of the Histochemistry Society
    |February 1, 1975
    PubMed
    Summary

Calcium is found in platelet alpha-granules and rapidly disappears upon thrombin stimulation. This suggests a key regulatory role for this calcium pool in platelet activation and response coupling.

    Area of Science:

    • Biochemistry
    • Cell Biology
    • Hematology

    Background:

    • Platelets play a crucial role in hemostasis and thrombosis.
    • Calcium ions are essential signaling molecules in cellular processes, including platelet activation.
    • The precise localization and dynamics of calcium within platelets are not fully understood.

    Purpose of the Study:

    • To investigate the localization of calcium within platelet alpha-granules.
    • To determine the dynamics of this calcium pool upon platelet stimulation.
    • To explore the potential regulatory role of alpha-granule calcium in platelet function.

    Main Methods:

    • Electron microscopic x-ray microprobe analysis was employed.
    • Pyroantimonate precipitation was used to localize calcium.

    Related Experiment Videos

  • Platelets were fixed with osmium tetroxide-pyroantimonate.
  • Platelet activation was induced by thrombin stimulation.
  • Main Results:

    • Calcium was localized within the nucleoids of platelet alpha-granules.
    • This calcium pool significantly decreased within 10 seconds of thrombin stimulation.
    • The rapid depletion of alpha-granule calcium was observed.

    Conclusions:

    • Platelet alpha-granules contain a readily accessible pool of calcium.
    • This calcium pool is rapidly mobilized upon platelet activation by thrombin.
    • Alpha-granule calcium likely plays a critical regulatory role in platelet stimulus-response coupling.
