Summary

This study found CD8-like molecules on human sperm from both fertile and subfertile men. Spermatozoa from subfertile patients showed different CD8-like molecule expression intensity compared to fertile donors.

  • To investigate differences in CD8-like molecule presence on human ejaculated spermatozoa between fertile donors and subfertile patients with leukocytospermia.
  • To characterize the expression and localization of CD8-like antigens on human spermatozoa.

  • Indirect and absorption ELISA (Enzyme-Linked Immunosorbent Assay).
  • Indirect immunoelectron microscopy (IEM) and indirect immunofluorescence (IIF).
  • Monoclonal antibody (MoAb) CD8 used for immunodetection.

  • CD8 immunopositive spermatozoa were detected in all studied samples.
  • Anti-human MoAb CD8 recognized a common epitope on normal human spermatozoa and fetal thymocytes.
  • Absorption experiments showed a significant decrease in MoAb CD8 reactivity when preincubated with spermatozoa.
  • CD8-like antigen determinants were localized in the acrosomal region, neck, and sperm-tail plasma membrane of normozoospermic samples.
  • Leukocytospermic samples exhibited similar CD8-like antigen localization but varied intensity compared to normozoospermic samples.

Conclusions:

  • Human spermatozoa exhibit heterogeneity in the presence, localization, and expression of CD8-like antigen determinants.
  • These findings expand the understanding of CD8-like antigen characteristics in spermatozoa from both fertile and subfertile men.
