Summary

Peroxisome proliferator-activated receptor alpha (PPAR-alpha) activators, like fibric acids, improve lipid profiles and cardiovascular health. These compounds reduce triglycerides and increase HDL-cholesterol, benefiting patients with heart disease risk factors.

PPAR research·2017

Area of Science:

  • Molecular biology
  • Cardiovascular medicine
  • Metabolic research

Background:

  • PPAR-alpha is a nuclear receptor regulating genes in fatty acid and lipoprotein metabolism.
  • PPAR-alpha activators influence lipid concentrations and cardiovascular markers.

Purpose of the Study:

  • To summarize the metabolic and cardiovascular effects of PPAR-alpha activation.
  • To highlight the therapeutic potential of PPAR-alpha activators in managing dyslipidemia and related conditions.

Main Methods:

  • Review of studies on PPAR-alpha activators, including fibric acids like gemfibrozil, bezafibrate, and fenofibrate.
  • Analysis of clinical trial data (e.g., VA-HIT, BECAIT, DAIS) assessing cardiovascular outcomes.

Main Results:

Related Experiment Videos

  • PPAR-alpha activation decreases triglyceride levels by upregulating lipoprotein lipase and downregulating apo C-III.
  • HDL-cholesterol is increased via enhanced expression of apo A-I and apo A-II.
  • Activation improves insulin sensitivity, reduces thrombosis, and lessens vascular inflammation.

Conclusions:

  • PPAR-alpha activators demonstrate significant benefits in lipid management and cardiovascular risk reduction.
  • Clinical trials support the use of PPAR-alpha activators in specific patient populations, including those with low HDL-cholesterol and type 2 diabetes.
