Summary

Glucocorticoids regulate glutamine synthetase (GS) in female rat muscle. Sex steroids like progesterone and estradiol do not influence GS activity with aging, suggesting reproductive status doesn't impact muscle GS.

  • Glutamine synthetase (GS) is crucial for glutamine synthesis.
  • GS is induced by glucocorticoids and maintained during aging in skeletal muscle.
  • The role of sex steroids (progesterone, estradiol) in regulating muscle GS during aging is unknown.

Purpose of the Study:

  • To investigate the role of progesterone and estradiol in regulating glutamine synthetase (GS) with aging in female rats.
  • To determine if sex steroids influence muscle GS activity and mRNA expression in young and aged female rats.

Main Methods:

  • Adult and aged female Wistar rats underwent adrenalectomy (ADX), ovariectomy (OV), or both (ADXOV), compared to sham controls.
  • Muscle GS activity and mRNA expression in tibialis anterior were measured.

  • Hormone replacement therapy with progesterone and estradiol was assessed.

    • Adrenalectomy significantly decreased muscle GS activity and expression in both age groups.

    • Heart GS activity decreased with ADX only in aged rats.

    Conclusions:

    • Reproductive endocrine status does not affect skeletal muscle glutamine synthetase activity in female rats.
    • Aging may alter the regulation of heart glutamine synthetase by steroids.
