    Purpose of the Study:

    • To investigate cell-mediated immunity in human malignancies using the leukocyte migration test (L.M.T.).
    • To determine if localized tumors are recognized as antigenic by autologous leukocytes.

    Main Methods:

    • In vitro analysis of peripheral blood leukocytes and tumor cells from 35 untreated cancer patients.
    • Application of the leukocyte migration test (L.M.T.) with fresh, frozen-thawed, or extracted tumor cells.

    Main Results:

    • Leukocyte migration was inhibited in 18/20 patients with localized, resectable tumors, suggesting an antigenic response.
    • Leukocyte migration was not inhibited in 14/15 patients with metastatic or widespread disease.
    • Similar results were observed using fresh, frozen-thawed, or tumor cell extracts.

    Conclusions:

    • Localized tumors appear to be recognized as antigenic by presensitized autologous leukocytes, leading to migration inhibition.
    • This immune recognition and inhibition appear to be lost in the presence of hematogenous metastasis.
    • Normal tissues did not elicit an inhibitory response, indicating specific recognition of tumor-associated antigens.
