Summary

Adenovirus-mediated gene transfer is feasible for vein grafts. Transferring the prourokinase (Pro-UK) gene into vein grafts effectively prevented thrombosis and maintained graft patency.

Purpose of the Study:

  • To assess the feasibility of adenovirus-mediated gene transfer into vein grafts.
  • To investigate the protective role of the prourokinase (Pro-UK) gene against vein graft thrombosis.

Main Methods:

  • Adenovirus vectors (Adv5-CMV, Adv5-CMV/LacZ, Adv5-CMV/Pro-UK) were used for ex vivo gene transfer into rat jugular vein grafts.
  • Grafts were implanted as interposition conduits in the carotid arteries.
  • Platelet uptake, gene expression (beta-galactosidase, Pro-UK), thrombosis, and occlusion rates were evaluated.
