Summary

Late pregnancy reduces blood pressure by altering voltage-dependent calcium channels (VDCC). Activating VDCC in pregnant rats increased blood pressure, while blocking VDCC in nonpregnant rats decreased it.

Background:

  • Pregnancy induces significant hemodynamic changes, including decreased vascular resistance and blood pressure.
  • Altered activity of voltage-dependent calcium channels (VDCC) in the adrenal cortex and vascular smooth muscle during late pregnancy is linked to reduced blood pressure.

Purpose of the Study:

  • To investigate the role of voltage-dependent calcium channels (VDCC) in pregnancy-induced hypotension.
  • To determine if modulating VDCC activity could prevent or mimic pregnancy-related blood pressure changes.

Main Methods:

  • Pregnant rats were treated with a VDCC activator (CGP 28392) to prevent hypotension.
  • Nonpregnant rats received a VDCC blocker (nifedipine) to mimic pregnancy-induced hypotension.

    • CGP 28392 treatment in pregnant rats caused a transient increase in systolic blood pressure, which then declined but remained higher than controls.

    • Neither treatment affected body weight, intake, plasma renin activity, aldosterone, or corticosterone levels.

    Conclusions:

    • Voltage-dependent calcium channel (VDCC) activation can induce hypertension.
    • Modulating VDCC function influences the extent of blood pressure decrease observed at the end of pregnancy.
    • These findings highlight the critical role of VDCC in regulating blood pressure during gestation.
