Area of Science:

  • Mycology
  • Biochemistry
  • Immunology

Background:

  • Ganoderma lucidum (Lingzhi mushroom) is traditionally used for health benefits.
  • Polysaccharides are bioactive compounds found in mushrooms with potential immunomodulatory effects.

Purpose of the Study:

  • To isolate and characterize polysaccharides from Ganoderma lucidum fruit bodies.
  • To investigate the in vitro and in vivo immunomodulatory activities of these polysaccharides.

Main Methods:

  • Isolation using anion-exchange and gel-filtration chromatography.
  • Structural elucidation via glycosyl composition, linkage analysis, NMR spectroscopy, and mass spectrometry.
  • In vitro lymphocyte proliferation assays and in vivo immune stimulation tests in mice.

Main Results:

  • Three polysaccharides (PL-1, PL-3, PL-4) were isolated and their complex structures determined.
  • PL-1, PL-3, and PL-4 enhanced T- and B-lymphocyte proliferation in vitro.
  • PL-1 demonstrated significant immune-stimulating activity in mice.

Conclusions:

  • Ganoderma lucidum fruit bodies contain structurally diverse polysaccharides with immunomodulatory properties.
  • These polysaccharides, particularly PL-1, show potential as immune-enhancing agents.
