I.R.-Stimulated catalysis (a potential spectral tool).

T F Smith, M Ross

International Journal of Peptide and Protein Research

    Summary

Infrared spectrography may offer a new index for catalytic rate. A mutant beta-galactosidase showed a 1-2 fold increase in activity when exposed to 10.6 micrometer irradiation, unlike the wild-type enzyme.

    Area of Science:

    • Biochemistry
    • Enzymology
    • Spectroscopy

    Background:

    • Catalytic rate is a key enzyme property.
    • Infrared spectroscopy is a non-invasive analytical technique.

    Purpose of the Study:

    • To investigate infrared spectrography as an index for catalytic activity.
    • To assess the effect of specific infrared irradiation on enzyme kinetics.

    Main Methods:

    • Enzyme kinetics assays were performed on beta-galactosidase.
    • A mutant and wild-type enzyme were exposed to 10.6 micrometer infrared irradiation.
    • Catalytic rates were measured and compared.

    Main Results:

    • A 1-2 fold increase in catalytic rate was observed for the mutant enzyme.
    • No significant change in catalytic rate was observed for the wild-type enzyme under the same conditions.

    Conclusions:

    • Infrared irradiation at 10.6 micrometers may influence the catalytic activity of specific enzyme mutants.
    • Catalytic rate could potentially be indexed using infrared spectrographic methods, particularly for mutant enzymes.
