Quetiapine treatment prevented stress-induced reductions in brain-derived neurotrophic factor (BDNF) in rat hippocampus. This suggests quetiapine may offer neuroprotection for hippocampal neurons in schizophrenia.

Area of Science:

  • Neuroscience
  • Pharmacology
  • Molecular Biology

Background:

  • Schizophrenia is a psychotic disorder often associated with altered neurotrophic factor levels.
  • Chronic stress can negatively impact neuronal health, particularly in the hippocampus.
  • Brain-derived neurotrophic factor (BDNF) plays a crucial role in neuronal survival and function.

Purpose of the Study:

  • To investigate the effect of quetiapine on stress-induced changes in BDNF expression.
  • To determine if quetiapine exhibits neuroprotective properties in the hippocampus under stress conditions.

Main Methods:

  • Rats were subjected to chronic immobilization stress.
  • Quetiapine pretreatment (10 mg/kg) was administered.
  • BDNF protein levels were quantified using Western blot analysis.

Related Experiment Videos

  • BDNF immunoreactivity in hippocampal neurons was assessed.
  • Main Results:

    • Quetiapine pretreatment significantly attenuated the stress-induced decrease in hippocampal BDNF protein levels.
    • The reduction in BDNF immunoreactivity in hippocampal pyramidal and dentate granular neurons was also mitigated by quetiapine.
    • These findings indicate a neuroprotective effect of quetiapine against stress-induced neuronal changes.

    Conclusions:

    • Chronic quetiapine administration may protect hippocampal neurons from stress-induced damage.
    • The neuroprotective effect of quetiapine could be linked to its antipsychotic efficacy in schizophrenia patients.
