Summary

Odorant receptors (ORs) determine how olfactory sensory neurons (OSNs) respond to specific odors. This study shows ORs guide neuron connections in the olfactory bulb (OB), forming functional units.

Area of Science:

  • Neuroscience
  • Olfactory system research
  • Molecular biology

Background:

  • Odorant receptors (ORs) on olfactory sensory neurons (OSNs) detect odorants.
  • OSNs expressing the same OR converge to specific glomeruli in the olfactory bulb (OB).
  • The functional similarity of OSNs expressing a defined OR has not been experimentally verified.

Purpose of the Study:

  • To experimentally test the functional similarity of OSNs expressing a defined OR.
  • To investigate the role of ORs in determining odorant specificity and axonal convergence.
  • To understand the molecular basis of glomerular formation in the OB.

Main Methods:

  • Utilized calcium imaging in gene-targeted mice coexpressing green fluorescent protein with specific ORs.
  • Characterized odorant response properties of single OSNs.

Related Experiment Videos

  • Genetically modified OSNs by replacing OR coding sequences.
  • Main Results:

    • Demonstrated that acetophenone and benzaldehyde are agonists for the M71 OR.
    • Showed M71-expressing neurons exhibit functionally similar response properties across concentrations.
    • Confirmed that replacing the M71 OR with the rat I7 OR altered OSN response profiles and OB glomerular structure.
    • Established that the mouse I7 OR confers a specific response profile irrespective of epithelial expression zone.

    Conclusions:

    • ORs dictate both odorant specificity and axonal convergence.
    • OR expression is the molecular basis for the formation and arrangement of functional glomerular units in the OB.
    • This study provides direct evidence linking OR identity to functional OSN properties and OB organization.
