Children with oppositional defiant disorder (ODD) show heightened sensitivity in their postsynaptic serotonin 5-HT(1B/1D) receptors. This heightened sensitivity may be linked to impulsive aggression and altered stress responses.

Area of Science:

  • Neuroscience
  • Child Psychiatry
  • Pharmacology

Background:

  • Central serotonergic function disturbances are linked to impulsive aggression.
  • Specific serotonin 5-HT(1B) receptors are implicated in impulsive aggressive behavior.
  • Oppositional defiant disorder (ODD) in children warrants investigation into receptor functioning.

Purpose of the Study:

  • To investigate serotonin 5-HT(1B/1D) receptor functioning in children diagnosed with ODD.
  • To explore the relationship between receptor function and impulsive aggression in this population.

Main Methods:

  • Examined growth hormone (GH) response to sumatriptan (a 5-HT(1B/1D) agonist) in 20 children with ODD and 15 normal controls (NC).
  • Assessed stress response by measuring salivary cortisol levels.

Related Experiment Videos

  • GH and cortisol levels were monitored following sumatriptan administration.
  • Main Results:

    • Children with ODD exhibited a significantly stronger GH response to sumatriptan compared to NC.
    • NC children showed a significant cortisol increase post-injection, indicating a stress response.
    • The ODD group did not display a similar cortisol increase, suggesting altered stress reactivity.

    Conclusions:

    • The findings suggest enhanced functional sensitivity of postsynaptic 5-HT(1B/1D) receptors in children with ODD.
    • This heightened receptor sensitivity may contribute to the neurobiological underpinnings of ODD and associated behaviors.
