Structural analysis of large macrocycles, cyclotri(deoxycholate) and cyclotetra(24-norcholate), was achieved using X-ray crystallography and AM1 calculations. These methods revealed both solid-state and gas-phase molecular structures.
Area of Science:
- Supramolecular Chemistry
- Structural Biology
- Computational Chemistry
Background:
- Understanding the three-dimensional structures of complex molecules is crucial in chemistry and biology.
- Macrocycles, particularly large open ones, present unique structural challenges due to their conformational flexibility.
Purpose of the Study:
- To determine the solid-state and gas-phase structures of cyclotri(deoxycholate) and cyclotetra(24-norcholate).
- To investigate the structural characteristics of large open macrocycles.
Main Methods:
- X-ray crystallographic determinations were employed to elucidate solid-state structures.
- Austin Model 1 (AM1) calculations were utilized for gas-phase structure analysis.
Main Results:
- The solid-state and gas-phase structures of cyclotri(deoxycholate) were successfully defined.
- The solid-state and gas-phase structures of cyclotetra(24-norcholate) were determined.
- Cyclotetra(24-norcholate) represents one of the largest open macrocycles ever characterized by crystallography.
Conclusions:
- The study successfully defined the structures of two significant macrocycles.
- The findings contribute to the understanding of large macrocycle structural chemistry.

Ionic Crystal Structures
Most monatomic ions behave as charged spheres, and their attraction for ions of opposite charge is the same in every direction. Consequently, stable structures for ionic compounds result (1) when ions of one charge are surrounded by as many ions as possible of the opposite...
Acid Strength and Molecular Structure
In the absence of any leveling effect, the acid strength of binary compounds of hydrogen with nonmetals (A) increases as the H-A bond strength decreases down a group in the periodic table. For group 17, the order of increasing acidity is HF < HCl < HBr < HI. Likewise, for group 16, the order of increasing acid strength is H2O < H2S < H2Se < H2Te. Across a row in the periodic table, the acid strength of binary hydrogen compounds increases with increasing...
Structural Isomerism
Isomers are different chemical species that have the same chemical formula. Structural isomerism of coordination compounds can be divided into two subcategories, the linkage isomers and coordination-sphere isomers.
Linkage isomers occur when the coordination compound contains a ligand that can bind to the transition metal center through two different atoms. For example, the CN− ligand can bind through the carbon atom or through the nitrogen atom. Similarly, SCN− can be...
Chirality at Nitrogen, Phosphorus, and Sulfur
A consequence of chirality is the need for enantiomeric resolution. While this is theoretically possible for all...
Structures of Carboxylic Acid Derivatives
Carboxylic acid derivatives contain an acyl group attached to a heteroatom such as chlorine, oxygen, or nitrogen. The carbonyl carbon and oxygen are both sp2-hybridized with an unhybridized p orbital.
The three sp2 orbitals of the carbonyl carbon form three σ bonds, one each with the carbonyl oxygen, the α carbon, and the heteroatom, whereas the other two sp2 orbitals of the carbonyl oxygen are occupied by the lone pairs. Further, the unhybridized p...
