Summary

The alpha(2) adrenoceptor plays a role in anesthesia. This study shows that the alpha(2) agonist clonidine increased propofol-induced sleep time, while the alpha(2) antagonist yohimbine decreased it, affecting norepinephrine release.

Background:

Purpose of the Study:

  • To investigate the effects of clonidine and yohimbine on propofol-induced sleep time in rats.
  • To correlate these effects with changes in prefrontal cortex norepinephrine release.

Main Methods:

  • Male Wistar rats were administered varying doses of yohimbine or clonidine, followed by propofol.
  • Sleep duration and prefrontal cortex norepinephrine levels (via microdialysis) were measured.

  • Yohimbine (1 mg/kg) decreased propofol sleep time by ~30% and increased norepinephrine by ~240%.
