Next-generation Sequencing03:00

Next-generation Sequencing

The first human genome sequencing project cost $2.7 billion and was declared complete in 2003, after 15 years of international cooperation and collaboration between several research teams and funding agencies. Today, with the advent of next-generation sequencing technologies, the cost and time of sequencing a human genome have dropped over 100 fold.
Next-Generation Sequencing Methods
Although all next-generation methods use different technologies, they all share a set of standard features.

Area of Science:

  • Genomics
  • Bioinformatics

Background:

  • Maintaining high sample throughput in DNA sequencing is crucial.
  • Network dependency can cause significant downtime for sequencing instruments.

Purpose of the Study:

  • To develop a novel software tool, Loader Lite, for the ABI PRISM 3700 DNA sequencer.
  • To enable autonomous operation of sequencing instruments, ensuring continuous sample processing.

Main Methods:

  • Loader Lite generates plate records using anonymous tray barcodes at runtime.
  • Barcode data is reconciled with project details via a database post-sequencing.
  • Plate records are created locally on a computer serving an individual sequencer before data import.

Main Results:

  • Loader Lite facilitates instrument operation independent of external networks.
  • Real-time plate record synthesis at the point of loading minimizes sample loading errors.
  • The software supports partial or full 384-well sample trays with configurable run parameters.

Conclusions:

  • Loader Lite enhances the operational efficiency and reliability of the ABI PRISM 3700 DNA sequencer.
  • The software's ability to prevent downtime due to network issues is a significant advantage.
  • Loader Lite has been successfully used for millions of sequencing reactions, indicating its utility for the broader scientific community.
