Area of Science:

  • Immunology
  • Cell Therapy
  • Molecular Biology

Background:

  • Adoptive T-cell therapies aim to enhance immune responses against diseases.
  • Current strategies seek to combine antibody specificity with immune cell effector functions.
  • Antigen-selective cell therapies are being developed for MHC- and cytokine-independent T-cell activation.

Purpose of the Study:

  • To review innovative strategies in adoptive T-cell therapy, focusing on antigen-selective approaches.
  • To discuss the development and potential of chimeric immune receptors (CIRs) and biespecic antibody-mediated immune recruitment (BIR).
  • To explore the transition from effector cell-based to progenitor cell-based therapies for sustained immunity.

Main Methods:

  • Genetic manipulation of T-cells to express chimeric immune receptors (CIRs) with antibody specificity.

Related Experiment Videos

  • Utilizing biespecic antibody fragments for trans-recruitment and multieffector activation.
  • Evaluating both in vitro and in vivo model systems for therapeutic efficacy.
  • Main Results:

    • CIR-expressing cells demonstrate specific stimulation and antigen-bearing cell killing.
    • Progenitor cell-based therapies can generate long-term persistent systemic immunity.
    • Biespecic antibody-mediated immune recruitment (BIR) approaches show promise in complementing CIR therapies.

    Conclusions:

    • Antigen-selective cell therapies, including CIR and BIR, represent significant advancements in cell-mediated immunotherapy.
    • Further research is needed to optimize effector cell selection and targeting strategies.
    • Ongoing clinical trials will determine the efficacy of these novel immunotherapies in cancer and HIV patients.
