Area of Science:

  • Pharmacology
  • Cardiovascular Medicine
  • Drug Development

Background:

  • BIBR-1048 is an orally-active prodrug designed as a thrombin inhibitor.
  • It is a prodrug of BIBR-953ZW, developed by Boehringer Ingelheim.
  • The compound is being investigated as a potential antithrombotic agent.

Purpose of the Study:

  • To evaluate BIBR-1048 as a novel antithrombotic therapy.
  • To assess its efficacy in preventing thromboembolism and stroke.
  • To demonstrate proof-of-principle in clinical trials for deep vein thrombosis.

Main Methods:

  • Phase II clinical trials were conducted to assess the efficacy and safety of BIBR-1048.
  • The drug was investigated for indications including thromboembolism, stroke prevention in atrial fibrillation, and deep vein thrombosis.

Main Results:

  • Proof-of-principle was demonstrated in phase II trials for deep vein thrombosis by April 2002.
  • Phase II trials were ongoing by 1999 for thromboembolism and stroke prevention in atrial fibrillation.

Conclusions:

  • BIBR-1048 shows promise as an orally-administered antithrombotic agent.
  • Clinical development has progressed through phase II trials, indicating potential therapeutic value.
  • Further development, including IND filing for BIBR-953ZW, was anticipated.
