Summary

HER-2/neu protein overexpression in germ cell tumors is not reliably detected by immunohistochemistry (IHC) compared to fluorescence in situ hybridization (FISH). This lack of correlation suggests limited prognostic or therapeutic value for HER-2/neu in these cancers.

Background:

Purpose of the Study:

  • To evaluate HER-2/neu positivity rates in GCTs using both immunohistochemistry (IHC) and fluorescence in situ hybridization (FISH).
  • To determine the concordance between IHC and FISH results for HER-2/neu assessment in GCTs.
  • To ascertain the potential prognostic and therapeutic implications of HER-2/neu in GCTs.

Main Methods:

  • Analysis of 96 archival GCT pathology specimens.
  • HER-2/neu protein expression assessed by IHC using the HercepTest kit.
  • HER-2/neu gene amplification evaluated by FISH with the INFORM assay and a centromere 17 probe.

  • Twenty-two out of 96 specimens (22.9%) showed HER-2/neu protein overexpression by IHC.
