Summary
Murine embryonic stem cells generate functional megakaryocytes for studying alphaIIbbeta3 integrin signaling. CalDAG-GEFI enhances agonist-induced fibrinogen binding in these cells.
Area of Science:
- Hematology
- Cell Biology
- Molecular Biology
Background:
- Integrin alphaIIbbeta3 mediates platelet aggregation via fibrinogen binding, regulated by inside-out signaling.
- Megakaryocytes, platelet precursors, also exhibit agonist regulation of alphaIIbbeta3.
- Understanding inside-out signaling in megakaryocytes is crucial for platelet function research.
Purpose of the Study:
- To generate functional megakaryocytes from murine embryonic stem (ES) cells for studying alphaIIbbeta3 integrin signaling.
- To investigate the role of CalDAG-GEFI in agonist-induced alphaIIbbeta3 activation in ES cell-derived megakaryocytes.
Main Methods:
- Coculture of murine ES cells with OP9 stromal cells and specific cytokines (thrombopoietin, IL-6, IL-11) for megakaryocyte differentiation.
- Analysis of megakaryocyte markers and proplatelet formation.
- Functional assays measuring fibrinogen binding to alphaIIbbeta3 in response to agonists.
- Retrovirus-mediated gene expression of green fluorescent protein and CalDAG-GEFI.
Main Results:
- ES cells differentiated into mature, polyploid megakaryocytes expressing alphaIIbbeta3 and producing proplatelets.
- Mature megakaryocytes, but not progenitors, showed agonist-dependent fibrinogen binding via alphaIIbbeta3.
- Retroviral expression of CalDAG-GEFI enhanced agonist-induced Rap1b activation and alphaIIbbeta3-mediated fibrinogen binding.
- Reporter gene expression did not impair megakaryocyte viability or alphaIIbbeta3 function.
Conclusions:
- Murine ES cells provide a valuable source of mature megakaryocytes for research, including integrin signaling studies.
- CalDAG-GEFI plays a significant role in the inside-out signaling pathway regulating alphaIIbbeta3 function.
- This model system facilitates mechanistic studies of integrin regulation and platelet biology.

Mesenchymal Stem Cells
Mesenchymal stem cells (MSCs) are adult stem cells that can differentiate into most connective tissue cell types, except for hematopoietic cells, depending upon the source of MSCs. For example, bone-marrow-derived MSCs (BM-MSCs) can differentiate into osteocytes, hepatocytes, and pancreatic and neuronal cells. MSCs can be isolated from various sources such as bone marrow, placenta, adipose tissue, teeth, and Wharton’s jelly, a gelatinous substance in the umbilical cord. The ease of their access...
Integrins
Animal and protozoan cells do not have cell walls to help maintain shape and provide structural stability. Instead, these eukaryotic cells secrete a sticky mass of carbohydrates and proteins into the spaces between adjacent cells. This network of proteins and molecules is called an extracellular matrix or ECM.
Some ECM proteins assemble into a basement membrane to which the remaining components adhere. Proteoglycans typically form the bulk of the ECM while fibrous proteins, like collagen,...

Activation of Integrins
Integrins bind ligands and transmit information from outside the cell to inside or vice-versa through an "outside-in signaling" or "inside-out signaling."
In "outside-in signaling," external factors in the extracellular space bind to exposed ligand binding sites on integrins. This causes the inactive protein to undergo a conformational change to become active. Integrins are often clustered on the cell membrane. Repetitive and regularly spaced ligand binding events provide an effective stimulus.

Intracellular Signaling Affects Focal Adhesions
Integrins act both as extracellular input receivers and as intracellular processing activators. As their name suggests, integrins are entirely integrated into the membrane structure. Their hydrophobic membrane-spanning regions interact with the phospholipid bilayer's hydrophobic region. These membrane receptors provide extracellular attachment sites for effectors like hormones and growth factors. They activate intracellular response cascades when their effectors are bound and active.

Immunoglobulin-like Cell Adhesion Molecules
Immunoglobulin-like cell adhesion molecules or Ig-CAMs are a versatile group of cell surface glycoproteins belonging to the immunoglobulin protein superfamily. Ig-CAMs possess the characteristic immunoglobulin protein domains and other domains such as the fibronectin type III domain. The Ig domains are glycosylated to varying degrees in different Ig-CAMs.
Ig-CAMs exhibit either homophilic binding (to other Ig-CAMs) or heterophilic binding (to other ligands such as integrins). While most Ig-CAMs...
