    Guanacline treatment can cause persistent parotid pain, potentially due to sympathetic nerve damage. This study compared patients with and without this side effect, finding a link to higher doses and other adverse effects.

    Background:

    • Guanacline is an antihypertensive medication.
    • Parotid pain is a reported side effect of guanacline therapy.
    • The underlying mechanism of guanacline-induced parotid pain is not fully understood.

    Purpose of the Study:

    • To investigate the characteristics of patients who developed parotid pain during guanacline treatment.
    • To compare patients with parotid pain to those without this side effect.
    • To explore potential causes of persistent parotid pain after guanacline cessation.

    Main Methods:

    • A comparative study of 17 patients with guanacline-induced parotid pain and 17 control patients.
    • Analysis of patient demographics, clinical diagnoses, laboratory values, and concurrent medications.
    • Assessment of the association between parotid pain and other side effects or guanacline dosage.

    Main Results:

    • No significant differences were found in sex, age, diagnosis, blood urea, or prior hypotensive therapy between groups.
    • Parotid pain showed a slight association with other guanacline side effects.

    • Persistent parotid pain after guanacline therapy may result from prolonged or permanent sympathetic nerve damage.
    • The damage might affect the cervical sympathetic nerve supply to the salivary glands.
    • Further research is needed to confirm the neurogenic etiology of this adverse effect.
