    Background:

    • Studying intact tissue cell surfaces with scanning electron microscopy (SEM) is challenging.

    Purpose of the Study:

    • To develop a novel method for examining all surfaces of differentiated cells from intact tissues using SEM.
    • To expose and visualize previously inaccessible cell topography.

    Main Methods:

    • Tissue fixation with OSO4 followed by boric acid treatment.
    • Induction of cell dissociation via mechanical pressure after specific soaking times.
    • Cell adhesion to a substrate and preparation for SEM (critical point drying).

    • Successfully visualized the complete topography of various cell types, including hepatocytes, intestinal absorptive cells, kidney proximal tubule cells, mammary tumor cells, and sarcoma cells.
    • Demonstrated the dissociation of several other tissue types using modified procedures.

    • This technique provides new insights into cell topography and is applicable to a range of tissues with minor modifications.
