Short-chain fatty acid (SCFA) enemas, particularly butyrate, did not reduce colonic hypersensitivity in rats with colitis. Instead, butyrate enemas increased pain sensitivity and inflammation duration, questioning their therapeutic benefit for inflammatory bowel disease.

Background:

  • Short-chain fatty acids (SCFAs), especially butyrate, are commonly used in enemas for inflammatory bowel disease (IBD).
  • Their efficacy in reducing IBD symptoms is widely accepted, but direct evidence on colonic sensitivity is limited.

Purpose of the Study:

  • To investigate the actual effects of SCFA enemas on colonic sensitivity in rats.
  • To evaluate the impact of butyrate and other SCFAs on visceral pain thresholds in both healthy and colitis-induced rats.

Main Methods:

  • Rats were treated with enemas of saline, acetate, propionate, or butyrate.
  • Visceral pain thresholds were measured using colonic distension in control rats and rats with colitis induced by TNBS.
