Summary

Amiodarone significantly increases the risk of thyroid disorders, including hyperthyroidism and hypothyroidism. Higher cumulative doses of amiodarone correlate with a greater risk of developing these thyroid conditions.

Purpose of the Study:

  • To determine the risk of thyroid disorder in patients initiating amiodarone treatment.
  • To quantify the association between amiodarone use and the incidence of hyperthyroidism and hypothyroidism.

Main Methods:

  • A nested case-control study was conducted within a cohort of 5522 patients starting an anti-arrhythmic drug.
  • Cases were defined as patients developing hyperthyroidism or hypothyroidism after amiodarone initiation.

    • A dose-response relationship was observed, though less pronounced for hypothyroidism.

    Conclusions:

    • Amiodarone is associated with a notable increased risk of thyroid disorders.
    • Higher cumulative doses of amiodarone amplify the risk of developing thyroid dysfunction.
    • Clinicians must remain vigilant for thyroid disorders in patients treated with amiodarone, even with long-term use.
