Summary

Etanercept effectively reduces rheumatoid arthritis (RA) symptoms and structural damage, even in patients with inadequate responses to other treatments. Combination therapy with methotrexate is beneficial for severe cases, with a low discontinuation rate due to adverse events.

Area of Science:

  • Immunology
  • Rheumatology
  • Pharmacology

Background:

  • Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by joint inflammation and potential structural damage.
  • Tumor Necrosis Factor-alpha (TNF) plays a key role in the inflammatory process of RA.
  • Existing disease-modifying antirheumatic drugs (DMARDs) may not be effective for all RA patients.

Purpose of the Study:

  • To evaluate the efficacy and safety of Etanercept in managing active rheumatoid arthritis.
  • To assess the role of Etanercept in inhibiting structural damage in RA patients.
  • To explore the potential benefits of combination therapy with Etanercept and methotrexate.

Main Methods:

  • Etanercept, a biologic agent, targets TNF by acting as a soluble receptor fusion protein.
  • Clinical trials assessed patient responses in terms of signs, symptoms, and radiographic progression.
  • Safety profiles, including adverse events, serious infections, and malignancies, were monitored.

Main Results:

  • Etanercept demonstrated efficacy in reducing RA signs and symptoms and inhibiting structural damage progression.
  • Approximately 4% of patients discontinued therapy due to adverse events.
  • Etanercept did not significantly increase the risk of serious infections or malignancies.

Conclusions:

  • Etanercept is an effective treatment option for active RA patients with an inadequate response to DMARDs.
  • Combination therapy with methotrexate may be advantageous for severely active RA patients.
  • Etanercept offers a favorable safety profile, with manageable adverse events and no significant increase in serious risks.
