Summary

Yeast tRNA(Trp) processing is unique, with 3' maturation occurring before 5' cleavage, unlike other tRNAs. This atypical pathway still requires Lsm proteins but not Lhp1p for 3' maturation.

Area of Science:

  • Molecular Biology
  • RNA Processing
  • Eukaryotic Gene Expression

Background:

  • Eukaryotic pre-tRNA processing typically involves 5'-cleavage by RNase P before 3'-maturation.
  • The Lsm proteins and the La homolog Lhp1p are known to be essential for normal pre-tRNA processing and stability.

Purpose of the Study:

  • To investigate the processing pathway of yeast tRNA(Trp).
  • To understand the role of Lsm proteins and Lhp1p in the maturation of tRNA(Trp).

Main Methods:

  • Analysis of pre-tRNA processing intermediates.
  • Investigating the dependence on essential proteins like Lsm and Lhp1p.

Main Results:

  • Yeast tRNA(Trp) exhibits an unusual processing pathway where 3'-maturation precedes 5'-cleavage.
  • Pre-tRNA(Trp) processing depends on essential Lsm proteins for normal maturation and Lhp1p association.
  • Unlike other tRNAs, tRNA(Trp) does not require Lhp1p for its 3' processing and stability.

Conclusions:

  • Yeast tRNA(Trp) utilizes a distinct maturation pathway compared to other characterized tRNAs.
  • While Lhp1p is dispensable for tRNA(Trp) 3' processing, Lsm proteins remain crucial.
  • This finding highlights the diversity in tRNA processing mechanisms within eukaryotes.
