Macrophage migration inhibitory factor (MIF) is elevated in diffuse systemic sclerosis (dSSc) patients' skin and serum. This suggests MIF plays a role in the inflammatory processes driving scleroderma tissue remodeling.

  • Systemic sclerosis (SSc) is an autoimmune disease characterized by fibrosis and vascular abnormalities.
  • The diffuse form of SSc (dSSc) involves widespread skin thickening and internal organ involvement.
  • The role of macrophage migration inhibitory factor (MIF) in dSSc pathogenesis is not fully understood.

Purpose of the Study:

  • To investigate whether macrophage migration inhibitory factor (MIF) production is dysregulated in patients with diffuse systemic sclerosis (dSSc).

Main Methods:

  • The study included 10 dSSc patients and 10 age- and sex-matched healthy controls.
  • MIF expression was assessed in skin biopsies using immunohistochemistry.
  • MIF levels were quantified in serum and cultured fibroblast supernatants using ELISA.

  • MIF immunostaining was observed in keratinocytes and dermal fibroblasts in dSSc patients.
  • Serum MIF concentrations were significantly higher in dSSc patients compared to controls (p=0.011).

Conclusions:

  • MIF production appears to be dysregulated in patients with dSSc.
  • These findings suggest MIF may contribute to the pro-inflammatory loop involved in scleroderma.
  • MIF could play a role in the tissue remodeling characteristic of dSSc.
