Summary

Decorin (DCN) regulates bone mineralization. Higher DCN levels delay mineralization, while lower levels accelerate it, suggesting DCN inhibits collagen matrix mineralization timing.

Background:

  • Decorin (DCN) is a small leucine-rich proteoglycan.
  • DCN is known to modulate collagen fibrillogenesis.

Purpose of the Study:

  • To investigate the role of DCN in collagen matrix mineralization.
  • To characterize the effects of altered DCN levels on osteoblastic cells.

Main Methods:

  • Generated stable osteoblastic cell clones with higher (sense-DCN, S-DCN) and lower (antisense-DCN, As-DCN) DCN expression.
  • Characterized mineralized nodule formation in these cell clones.

Main Results:

  • Mineralization onset was significantly delayed in S-DCN clones compared to controls.
  • Mineralization was markedly accelerated and nodule number increased in As-DCN clones.
