Reference method for platelet enumeration.

Summary

Automated platelet counts using flow cytometry (FCM) with monoclonal antibodies show promise as a reference method. This study compared FCM results to electric resistance and optical methods for accurate platelet enumeration.

Background:

Automated hematology analyzers traditionally use electric resistance for platelet enumeration.
Optical systems are also increasingly used for platelet counting.
Flow cytometry (FCM) with monoclonal antibodies is being explored for accurate platelet enumeration.

Purpose of the Study:

To evaluate the International Society for Laboratory Hematology (ISLH) proposed monoclonal antibody-employed FCM method for platelet counting.
To compare FCM-based platelet counts with those from electric resistance and optical automated analyzers.

Main Methods:

Platelet counts were determined using the ISLH-recommended monoclonal antibody-employed FCM method.
Results were compared against counts from a Sysmex XE-2100 analyzer (electric resistance) and an optical system analyzer.

Main Results:

The study compared platelet counts obtained via FCM against established automated methods.
Analysis focused on the accuracy and reliability of the FCM approach for platelet enumeration.

Conclusions:

The monoclonal antibody-employed FCM method offers a potential reference standard for platelet counting.
Further validation is needed to establish FCM as a routine diagnostic tool for platelet enumeration.
