Background:

  • Systemic lupus erythematosus (SLE) is a multi-organ autoimmune disease characterized by anti-DNA antibodies.
  • Current broad-spectrum therapies (steroids, cytotoxic agents) have significant side effects.
  • There is a need for targeted therapies that address specific mechanisms of inflammation and autoimmunity in SLE.

Purpose of the Study:

  • To review recent advances in targeted molecular therapies for SLE.
  • To evaluate novel treatment modalities including immunosuppression and immune response modulation.
  • To identify promising therapeutic agents based on controlled and open trials.

Main Methods:

  • Literature review of MEDLINE search results from 2000-2002.
  • Inclusion of controlled trials, open trials, and Phase I/II trials.
  • Exclusion of anecdotal reports to focus on evidence-based findings.

  • Mycophenolate mofetil (MMF) and LJP 394 represent major advances in SLE treatment.
