Area of Science:

  • Pharmacology
  • Gastroenterology
  • Rheumatology

Background:

  • Rofecoxib is a selective cyclooxygenase-2 (COX-2) inhibitor.
  • Established efficacy in osteoarthritis, rheumatoid arthritis, and acute pain management.
  • Approved in the US for osteoarthritis and acute pain.

Purpose of the Study:

  • To evaluate the gastrointestinal safety and efficacy of rofecoxib compared to NSAIDs.
  • To assess rofecoxib's therapeutic benefit and toxicity profile.

Main Methods:

  • Clinical trials comparing rofecoxib to standard NSAIDs and placebo.
  • Endoscopic assessment of gastrointestinal ulceration.

Main Results:

  • Significantly lower rates of endoscopically proven gastrointestinal ulceration with rofecoxib versus NSAIDs.

Related Experiment Videos

  • Ulceration rates with rofecoxib were comparable to placebo.
  • Rofecoxib demonstrated equivalent clinical benefit to standard NSAIDs.
  • Conclusions:

    • Rofecoxib offers a favorable gastrointestinal safety profile compared to traditional NSAIDs.
    • Provides effective pain relief and anti-inflammatory benefits with reduced toxicity.
    • A valuable therapeutic option for conditions requiring NSAID treatment.
