Investigating the Kit gene

Area of Science:

  • Molecular Biology
  • Developmental Biology
  • Genetics

Background:

  • The Kit gene encodes a receptor tyrosine kinase crucial for development of stem cells and their progeny.
  • Understanding Kit gene transcriptional regulation is vital for developmental processes.
  • Previous research has not elucidated the molecular basis for Kit gene regulation.

Purpose of the Study:

  • To investigate the transcriptional regulation of the Kit gene in hematopoietic cells and primordial germ cells (PGCs).
  • To identify regulatory elements controlling Kit gene expression during development.

Main Methods:

  • Identified 6 DNase I hypersensitive sites (HS1-HS6) in the mouse Kit gene's promoter and first intron.
  • Developed transgenic mouse lines expressing green fluorescent protein (GFP) under the control of Kit regulatory elements.

Related Experiment Videos

  • Assessed GFP expression patterns in PGCs and hematopoietic progenitors.
  • Main Results:

    • A construct with the Kit promoter and all 6 HS sites showed high expression in PGCs and hematopoietic progenitors.
    • The Kit promoter alone (HS1) drove low-level PGC expression but not hematopoietic expression.
    • The HS2 fragment was necessary for hematopoietic expression and enhanced PGC expression, acting as a common enhancer.
    • Optimal hematopoietic expression required additional elements HS3-HS6.

    Conclusions:

    • The Kit gene's transcriptional regulation involves multiple cis-regulatory elements within its promoter and first intron.
    • The HS2 element functions as a crucial enhancer, integrating signals for both PGC and hematopoietic stem cell development.
    • Distinct combinations of regulatory elements (HS1-HS6) dictate cell-type-specific Kit gene expression patterns.
