Summary

This study presents a validated rat model for severe limb ischemia, suitable for studying metabolic and angiogenic healing processes over 8 weeks. The model effectively reduces foot perfusion, aiding research into angio- and arteriogenesis.

Area of Science:

  • Vascular Biology
  • Ischemia Research
  • Animal Modeling

Background:

  • Limb ischemia requires robust animal models to study pathophysiology and therapeutic interventions.
  • Existing models may not adequately discriminate between metabolic and angiogenic processes.

Purpose of the Study:

  • To develop and validate a severe limb ischemia rat model.
  • To assess its suitability for longitudinal studies of metabolic and angiogenic healing.

Main Methods:

  • A two-stage surgical procedure was used to induce unilateral limb ischemia in Sprague-Dawley rats.
  • Evaluation involved transit-time flowmetry, laser Doppler perfusion imaging, microspheres, arteriography, and histology over 8 weeks.

Main Results:

Related Experiment Videos

  • The model demonstrated low mortality and was well-tolerated.
  • Foot skin perfusion was significantly reduced for up to 8 weeks.
  • Collateral vessel formation was observed by 2 weeks, with histologic signs of ischemia lasting up to 4 weeks.
  • Conclusions:

    • This severe limb ischemia rat model is well-suited for long-term studies.
    • It allows for the investigation of pathophysiology and healing mechanisms, including angiogenesis and arteriogenesis.
