    Background:

    • Alloxan and streptozotocin are diabetogenic agents causing beta-cell necrosis in rats.
    • Previous research indicated D-glucose and related sugars protect against alloxan toxicity.

    Purpose of the Study:

    • To evaluate the protective effects of various agents against streptozotocin-induced beta-cell toxicity.
    • To compare the protective mechanisms of agents against streptozotocin and alloxan.

    Main Methods:

    • Streptozotocin (60 mg/kg) was administered intravenously to fasted male rats.
    • Potential protective agents were infused intravenously before or after streptozotocin.
    • Protection was assessed by plasma glucose levels and islet histology.

    Main Results:

    • 3-O-methyl-D-glucose and 2-deoxy-D-glucose provided significant protection against streptozotocin toxicity.
    • Unlike alloxan, protection against streptozotocin by these agents was effective even when administered post-treatment.
    • 3-O-methyl-D-glucose is the sole carbohydrate offering protection against both agents.

    Conclusions:

    • 3-O-methyl-D-glucose and 2-deoxy-D-glucose exhibit distinct protective mechanisms against streptozotocin compared to alloxan.
    • Differences in beta-cytotoxic mechanisms between alloxan and streptozotocin are suggested.
