A new primitive neuroectodermal tumor cell line, KK-2, was established from an occipital lobe tumor. This cell line, characterized by specific immunocytochemical markers, offers a model for studying neuroectodermal tumor development.

Background:

  • Primitive neuroectodermal tumors (PNETs) are aggressive central nervous system neoplasms.
  • Establishing continuous cell lines is crucial for understanding tumor biology and developing therapies.

Observation:

  • A tumor from the occipital lobe of a 19-year-old patient was analyzed.
  • Histology revealed undifferentiated small, round cells lacking neuronal or glial differentiation, suggesting a PNET.
  • Tumor cells were cultured for 3 years, leading to the establishment of the continuous KK-2 cell line.

Findings:

  • The KK-2 cell line was transplantable to nude mice.
  • Immunocytochemistry detected neuron-specific enolase, protein gene product 9.5, vimentin, TUJ1, and NCAM.
  • Key markers for glial differentiation (GFAP, S-100) and mature neuronal markers (neurofilament, synaptophysin) were notably absent.
