Identification of a unique nuclear receptor for 9-cis retinoic acid

Abstract:
Two families of nuclear receptors (RAR and RXR) for retinoic acid have previously been identified, cloned, and shown to be intimately involved with receptor-mediated regulation of gene transcription. All-trans-retinoic acid (t-RA) has been known to be a high-affinity ligand for RAR but only a weak ligand for RXR, while the endogenous ligand for RXR was unknown. Now two laboratories have isolated and chemically characterized 9-cis-retinoic acid (c-RA) and have shown that it is a naturally occurring ligand for RXR. These results suggest that isomerization of t-RA into c-RA may play an important role in retinoic acid biology.
