Summary

A mouse chromobox gene, M33, was identified and found to be similar to Drosophila Polycomb (Pc). M33 expression correlates with cell proliferation, suggesting a role in cellular memory.

Area of Science:

  • Developmental Biology
  • Epigenetics
  • Molecular Biology

Background:

  • The Drosophila gene Polycomb (Pc) regulates gene expression and is involved in inheriting determined cell states.
  • Pc shares a conserved chromobox domain with Heterochromatin Protein 1 (HP1), a heterochromatin component.

Purpose of the Study:

  • To identify and characterize a mouse homolog of the Polycomb gene.
  • To investigate the expression patterns and potential function of the mouse M33 gene.

Main Methods:

  • Isolation of the mouse M33 gene using the Pc chromobox sequence.
  • Analysis of M33 protein sequence for conserved domains.
  • In situ hybridization and northern blot analysis to determine M33 expression patterns.
  • Comparison of M33 characteristics with Pc.

Main Results:

  • A novel mouse gene, M33, was isolated, encoding a protein with homology to Pc, including shared chromodomain and C-terminal regions.
  • M33 expression during late embryonic development parallels that of cyclin B, suggesting regulation by cellular proliferation.
  • M33 exhibits characteristics similar to Pc, indicating it is a Pc-like chromobox gene.

Conclusions:

  • M33 is a Pc-like chromobox gene in mice.
  • M33 expression is likely controlled by the rate of cellular proliferation.
  • The study suggests potential roles for M33 and Pc in cellular memory mechanisms.
