Summary

P-glycoprotein (P-gp) overexpression in brain tumors may cause chemotherapy resistance. P-gp in brain endothelial cells may also limit drug delivery to tumors, contributing to treatment failure.

Area of Science:

  • Neuro-oncology
  • Cancer Biology
  • Pharmacology

Background:

  • P-glycoprotein (P-gp) overexpression in cancer cells leads to multidrug resistance.
  • P-gp also hinders drug penetration across the blood-brain barrier.
  • P-gp in brain tumors could be a key factor in chemotherapy resistance.

Purpose of the Study:

  • To investigate the expression and localization of P-gp in human brain tumors.
  • To determine if P-gp contributes to drug resistance in brain tumors.

Main Methods:

  • Immunohistochemistry using a mouse monoclonal antibody against P-gp.
  • Analysis of P-gp expression in 49 diverse brain tumor samples.

Main Results:

  • P-gp was detected in tumor cells of high-grade glioblastomas and a meningeal sarcoma.
  • P-gp was consistently present in endothelial cells of low-grade primary brain tumors.
  • P-gp expression was variable or absent in endothelial cells of anaplastic tumors and brain metastases.

Conclusions:

  • P-gp expression in tumor cells and vasculature supports its role in brain tumor drug resistance.
  • Findings suggest P-gp contributes to chemotherapy resistance in specific brain tumor types.
