Summary

Regulators of G protein signaling 8 (RGS8) mRNA expression begins postnatally in Purkinje cells, peaking around day 21. RGS8 protein is detected in the cell body and dendrites by day 14.

Area of Science:

  • Neuroscience
  • Molecular Biology
  • Developmental Biology

Background:

  • Regulators of G protein signaling (RGS) proteins are crucial modulators of heterotrimeric G protein signaling pathways.
  • RGS8, a member of the B/R4 subfamily, is known for its specific expression in adult cerebellar Purkinje cells.

Purpose of the Study:

  • To investigate the spatiotemporal expression pattern of RGS8 mRNA during cerebellar development.
  • To compare the developmental expression of RGS8 with RGS7 in the neonatal cerebellum.

Main Methods:

  • In situ hybridization was employed to detect RGS8 and RGS7 mRNA expression.
  • Immunohistochemistry using anti-RGS8 antibody was performed to analyze RGS8 protein localization.

Main Results:

  • RGS8 mRNA expression was first detected on postnatal day 9 in Purkinje cells.
  • RGS8 mRNA levels increased significantly until postnatal day 21, with a subsequent decrease in adult levels.
  • RGS7 mRNA was detected in the neonatal cerebellum by day 7.
  • RGS8 protein was observed in the cell body and dendrites of differentiating Purkinje cells by day 14, excluding the nucleus.

Conclusions:

  • RGS8 exhibits a distinct developmental expression profile in Purkinje cells, commencing postnatally and peaking in the second week.
  • The findings provide insights into the developmental roles of RGS8 in cerebellar circuitry and Purkinje cell maturation.
