Summary

Parallel tubular structures (PTS) are found in most CD16+ lymphocytes, suggesting a key role in target cell lysis. These structures were less common in CD57+ and CD8+ cells, indicating CD16+ lymphocytes are more cytotoxic.

Background:

  • Lymphocytes with large granular lymphocyte (LGL) morphology can contain parallel tubular structures (PTS) and electron-dense granules.
  • These structures are hypothesized to contain molecules crucial for target cell lysis.

Purpose of the Study:

  • To investigate the presence of PTS in lymphocytes under various conditions.
  • To compare PTS presence in different lymphocyte subsets (CD16+, CD57+, CD8+).

Main Methods:

  • Comparison of PTS presence in freshly isolated and overnight-incubated peripheral blood lymphocytes.
  • Incubation with human pooled serum, Hodgkin's disease patient sera, and interferon-alpha.
  • Flow cytometry for immunophenotyping (CD16, CD57, CD8) and PTS detection.
