Summary

Cytokeratins 18 and 19 are not associated with human retinal pigment epithelial cell (HRPE) proliferation. Instead, these cytokeratins are markers of HRPE cell migration in vitro.

Background:

  • Human retinal pigment epithelial cells (HRPE) express cytokeratins 18 and 19.
  • Previous research suggested a link between cytokeratin expression and HRPE proliferation.

Purpose of the Study:

  • To investigate the role of cytokeratins 18 and 19 in the proliferation and migration of cultured HRPE.
  • To determine if cytokeratins 18 and 19 are markers of HRPE proliferation or migration.

Main Methods:

  • Immunohistochemical techniques were used to identify cell proliferation markers (bromodeoxyuridine, proliferating cell nuclear antigen) and cytokeratins.
  • Monoclonal antibodies RGE 53 and K4.62 were utilized for cytokeratin detection.
  • Micro-chemotaxis chambers assessed cell migration.

Main Results:

  • Cytokeratins 18 and 19 were expressed in a subset of HRPE cells.
  • HRPE cells expressing cytokeratins 18 and 19 were less likely to be proliferating.
  • Actively migrating HRPE cells consistently expressed cytokeratins 18 and 19, while stationary cells did not.

Conclusions:

  • Cytokeratins 18 and 19 are not markers of HRPE proliferation.
  • Cytokeratins 18 and 19 are involved in the in vitro migration of HRPE.
  • These cytokeratins may serve as indicators of simple epithelial cell migration in tissues.
