Summary

In vitro radiosensitivity assays, including MTT and clonogenic assays, were used to assess malignant glioma response to radiotherapy. While no direct correlation with survival was found, lower SF2 values indicated better outcomes in the MTT assay.

Background:

  • Radiotherapy is a primary treatment for malignant gliomas, significantly impacting survival.

Purpose of the Study:

  • To evaluate the correlation between in vitro radiosensitivity, measured by clonogenic and MTT assays, and patient survival in malignant gliomas.
  • To compare the efficacy of MTT and clonogenic assays in determining radiosensitivity.

  • Establishment of primary cultures from 39 out of 47 malignant glioma biopsies.
  • Determination of surviving fraction to 2 Gy (SF2) using MTT and clonogenic assays post-x-ray irradiation.

  • No direct SF2 correlation with survival, but MTT assay showed significantly lower SF2 in patients surviving >18 months (p=0.01).

Conclusions:

  • In vitro radiosensitivity assays, particularly MTT, show potential in predicting treatment response in malignant gliomas.
  • Further research is needed to establish a definitive link between SF2 values and long-term patient survival for radiotherapy optimization.
