    Background:

    • Benzoyl peroxide is a common ingredient in acne treatments.
    • Concerns exist regarding its potential for causing allergic contact dermatitis.

    Purpose of the Study:

    • To evaluate the contact sensitizing potential of different benzoyl peroxide gel formulations.
    • To determine if benzoic acid or related compounds share sensitizing properties.

    Main Methods:

    • Human maximization test was employed to assess sensitization.
    • Four benzoyl peroxide formulations (two 5% and two 10% gels) were tested.
    • Subjects who developed sensitization were re-challenged with all formulations.

    Main Results:

    • A high sensitization rate of 76% was observed in the study population.
    • All sensitized subjects exhibited allergic reactions to all four benzoyl peroxide formulations.
    • Benzoic acid and related compounds did not induce allergic reactions in sensitized individuals.

    Conclusions:

    • Benzoyl peroxide, in both 5% and 10% gel formulations, is a significant contact sensitizer.
    • The tested benzoic acid compounds do not appear to be responsible for the observed sensitization.
    • Clinical use of benzoyl peroxide may lead to a considerable number of allergic contact dermatitis cases.
