Area of Science:

  • Medicinal Chemistry
  • Immunology
  • Drug Discovery

Background:

  • Very late antigen-4 (VLA-4) is a key adhesion molecule involved in inflammatory and autoimmune diseases.
  • Targeting VLA-4 offers a therapeutic strategy for various immune-mediated conditions.
  • The discovery of a sulphonamide by-product with VLA-4 antagonistic activity initiated a new drug development program.

Purpose of the Study:

  • To present the synthesis of novel small molecule VLA-4 antagonists.
  • To detail the structure-activity relationship (SAR) studies conducted.
  • To report the in vivo evaluation of a selected lead compound.

Main Methods:

  • Chemical synthesis of a series of sulphonamide-based compounds.
  • In vitro assays to assess VLA-4 antagonistic activity.

Related Experiment Videos

  • Pharmacokinetic and pharmacodynamic studies in animal models.
  • Main Results:

    • Identification of a potent sulphonamide by-product with VLA-4 antagonistic properties.
    • Establishment of SAR for optimizing VLA-4 antagonist potency and selectivity.
    • Demonstration of in vivo efficacy of a selected small molecule antagonist.

    Conclusions:

    • Small molecule VLA-4 antagonists derived from a sulphonamide scaffold are feasible.
    • The developed compounds show promise for treating VLA-4 mediated diseases.
    • Further investigation into these antagonists is warranted for therapeutic development.
