T-cell receptor polymorphism in primary biliary cirrhosis

Background:

Purpose of the Study:
- To investigate the association between T-cell receptor constant beta-2 (TCRB2) chain polymorphism and primary biliary cirrhosis.
- To determine if TCRB2 polymorphism influences disease severity or progression in PBC patients.
Main Methods:
- Restriction fragment length polymorphism (RFLP) analysis was used to genotype TCRB2 in 70 PBC patients and 70 healthy controls.
- Polymerase chain reaction (PCR) amplification and Bgl II digestion were employed for RFLP analysis.
Main Results:
- A slight increase in heterozygous genotype frequency was observed in PBC patients (49%) compared to controls (40%).
- This increase was more pronounced in early-stage PBC patients (60% vs. 40%).
- Heterozygous patients exhibited a trend towards less severe disease, indicated by lower Mayo scores.
Conclusions:
- TCR constant beta-2 chain polymorphism does not appear to be a major factor in the development of primary biliary cirrhosis.
- The observed associations in early-stage disease warrant further investigation but suggest a limited role in overall disease pathogenesis.
