Area of Science:

  • Microbiology
  • Biochemistry
  • Bioinformatics

Background:

  • Metallo-beta-lactamases are enzymes that hydrolyze beta-lactam antibiotics.
  • Ambler Class B metallo-beta-lactamases are a diverse group with clinical significance.
  • Existing classification schemes may not fully capture the evolutionary relationships within this class.

Purpose of the Study:

  • To investigate the phylogenetic relationships among Ambler Class B metallo-beta-lactamases.
  • To propose a revised classification system for these enzymes based on evolutionary divergence.
  • To compare the evolutionary relatedness of Class B enzymes to other beta-lactamase classes.

Main Methods:

  • Phylogenetic analysis of amino acid sequences.
  • Structural alignment of enzyme structures.

Related Experiment Videos

  • Comparative analysis with serine beta-lactamase classes (A, C, D).
  • Main Results:

    • Ambler Class B metallo-beta-lactamases segregate into two distinct phylogenetic groups.
    • No significant sequence homology exists between members of different Class B groups.
    • Class B groups show less relatedness to each other than to serine beta-lactamase classes.

    Conclusions:

    • The current classification of Ambler Class B metallo-beta-lactamases requires revision.
    • A new phylogenetic classification scheme is proposed for Class B metallo-beta-lactamases.
    • Understanding these evolutionary relationships is crucial for antibiotic resistance research.
